Exploring the precision redox map during fasting-refeeding and satiation in C. elegans

Abstract Fasting is a popular dietary strategy because it grants numerous advantages, and redox regulation one mechanism involved. However, the precise changes with respect to species, organelles tissues remain unclear, which hinders understanding of metabolic mechanism, exploring precision map under various statuses great significance. Twelve redox-sensitive C. elegans strains stably expressing genetically encoded fluorescent probes (Hyperion sensing H 2 O Grx1-roGFP2 GSH/GSSG) in three (cytoplasm, mitochondria endoplasmic reticulum (ER)) were constructed two (body wall muscle neurons) confirmed respond challenge. The GSSG/GSH obtained by confocal microscopy during fasting, refeeding, satiation. We found that fasting condition, decreased most compartments, except for an increase mitochondria, while increased cytoplasm body ER neurons. After caused reversed In satiated state, markedly cytoplasm, neurons, exhibited no change muscle. Our study systematically precisely presents characteristics different states living animals provides basis further investigating metabolism optimizing guidance.
